Preliminary Study for Prediction of Eye Irritation Based on the European Union Risk Phrase Using the Hansen Solubility Parameter

The in vivo rabbit eye test (Draize test), which is listed in Organisation for Economic Co-operation and Development Test Guideline 405, has been to test the ocular toxicity of drugs. However, because of animal welfare issues, the sale of cosmetics subjected to Draize tests has been prohibited in the European Union (EU). Accordingly, in this study, we examined the possibility of predicting eye irritation based on EU risk phrase using the Hansen solubility parameter (HSP). Evaluation of test substances based on Hansen spheres showed that the accuracy, sensitivity, and specificity of this approach were 97.3%, 80.0%, and 100.0% for R41, respectively. Additionally, the accuracy, sensitivity, and specificity were 87.5%, 100.0%, and 77.8%, respectively, for R36. These results indicated that HSP values may be useful for predicting the EU risk phrase results with high reproducibility and could therefore have applications in evaluation of the safety of substances.
